The effect of carrier-mediated efflux of ciprofloxacin (Cip) on the susceptibility of Streptococcus pneumoniae to the drug was investigated. The minimum inhibitory concentration (MIC) of Cip was reduced for strains of S. pneumoniae R6 in the presence of sodium chloride. The MIC of the drug for S. pneumoniae R6, T-338, and D39 increased when glucose was added to the medium containing an erythromycin (Ery) non-susceptible strain of S. pneumoniae D39 or to S. pneumoniae R6 that had grown in a cefotaxime-containing medium, whereas addition of sorbitol did not alter the MICs of these strains. Although the Ery MIC for S. pneumoniae R6 was not affected by addition of glucose, the MIC for S. pneumoniae D39 was increased in the presence of 0.4% glucose, 0.4% sorbitol, or 2% sodium chloride. The MIC of cefotaxime for strains of S. pneumoniae R6 increased 2-3-fold in the presence of 1% sorbitol. These results indicate that carrier-mediated efflux of Cip reduces the MIC of the drug.Preparation and characterization of porous polymer monoliths for HPLC. A novel porous polymer monolith was prepared and used in HPLC. The monolith was prepared in situ with a mixture of styrene, divinylbenzene, and cross-linker 4,4'-(methylenebis(phenyl isocyanate))bisbenzoic acid. The monolith was then characterized by scanning electron microscopy, mercury intrusion porosimetry, nitrogen sorption analysis, and pore volume measurement. Separation of norepinephrine, propranolol, and caffeine was achieved on the monolith using the reverse phase chromatography technique. The column efficiency, retention, peak shape, and separation efficiency were measured and found to be satisfactory.
